Electric Stimulation of Astaxanthin Biosynthesis in Haematococcus pluvialis

چکیده

The green microalga Haematococcus pluvialis accumulates astaxanthin, a potent antioxidant pigment, as defense mechanism against environmental stresses. In this study, we investigated the technical feasibility of stress-based method for inducing astaxanthin biosynthesis in H. using electric stimulation two-chamber bioelectrochemical system. When cathodic (reduction) current 3 mA (voltage: 2 V) was applied to cells two days, considerable lysis and breakage algal were observed, possibly owing formation excess reactive oxygen species at cathode. Conversely, absence cell breakage, application anodic (oxidation) effectively stimulated voltage range 2–6 V, whereas same could not be induced untreated control. At an optimal 4 V (anodic current: 30 mA), content electro-treated h 36.9% higher than that cells. Our findings suggest treatment can used improve production culture if parameters, such strength duration, are regulated properly.

Antioxidant role of astaxanthin in the green alga Haematococcus pluvialis

The green unicellular alga, Haematococcus pluvialis has two antioxidative mechanisms against environmental oxidative stress: antioxidative enzymes in vegetative cells and the antioxidative ketocarotenoid, astaxanthin, in cyst cells. We added a reagent that generates superoxide anion radicals (O2 ), methyl viologen, to mature and immature cysts of H. pluvialis. Tolerance to methyl viologen was h...

Transformation of the green alga Haematococcus pluvialis with a phytoene desaturase for accelerated astaxanthin biosynthesis.

Astaxanthin is a high-value carotenoid which is used as a pigmentation source in fish aquaculture. Additionally, a beneficial role of astaxanthin as a food supplement for humans has been suggested. The unicellular alga Haematococcus pluvialis is a suitable biological source for astaxanthin production. In the context of the strong biotechnological relevance of H. pluvialis, we developed a geneti...

Hydrolysis kinetics of astaxanthin esters and stability of astaxanthin of Haematococcus pluvialis during saponification.

The reaction kinetics for the hydrolysis of astaxanthin esters and the degradation of astaxanthin during saponification of the pigment extract from the microalga Haematococcus pluvialis were investigated. Different concentrations of sodium hydroxide in methanol were used for the saponification under nitrogen in darkness at ambient temperature (22 degrees C) followed by the analysis of astaxanth...

Media Screening for Obtaining Haematococcus pluvialis Red Motile Macrozooids Rich in Astaxanthin and Fatty Acids

Astaxanthin from Haematococcus pluvialis is commercially produced in a two-stage process, involving green vegetative (macrozooid) and red aplanospore stages. This approach has been scaled up to an industrial process but constraints limit its commercial success and profitability, including: contamination issues, high pigment extraction costs, requirements for high light levels and photo-bleachin...

Transcriptome Analysis in Haematococcus pluvialis: Astaxanthin Induction by High Light with Acetate and Fe2+

Haematococcus pluvialis is a commercial microalga, that produces abundant levels of astaxanthin under stress conditions. Acetate and Fe2+ are reported to be important for astaxanthin accumulation in H. pluvialis. In order to study the synergistic effects of high light stress and these two factors, we obtained transcriptomes for four groups: high light irradiation (HL), addition of 25 mM acetate...
